Human Papillomavirus (HPV) belongs to the genus Papillomavirus of the family Papillomaviridae, which is a group of non-enveloped small double-stranded circular DNA viruses. Widely distributed in nature, HPV has strict host specificity and tissue tropism, mainly infecting human skin and mucosal epithelial cells. From an evolutionary perspective, the HPV genome is highly conserved, with high homology in core gene sequences among different subtypes, while also exhibiting certain genetic variations, which allows its classification into multiple genotypes. As a representative member of the Papillomaviridae family, the core biological characteristic of HPV is that its replication cycle is closely related to the differentiation cycle of host epithelial cells. The processes of viral genome replication, transcription and translation all depend on the differentiation state of host cells, and the viral genome can integrate into the host cell genome and exist stably.
HPV viral particles have an icosahedral symmetric structure with a diameter of approximately 52-55 nm. The overall structure consists of only two parts, a capsid and a core, without a lipid envelope structure. The viral capsid is composed of two structural proteins, namely the major capsid protein L1 and the minor capsid protein L2. Among them, the L1 protein is the main component of the capsid, forming pentameric structures through self-assembly. Seventy-two L1 pentamers are arranged in a T=7 symmetry to form the icosahedral outer shell of the virus. The L2 protein is present in a small amount, embedded in the gaps of the capsid formed by L1 proteins, and participates in the viral assembly and infection processes. The viral core is a double-stranded circular DNA genome with a length of approximately 8 kb. The genome can be divided into three parts: the early region (E region), the late region (L region) and the upstream regulatory region (URR). The early region contains multiple open reading frames, encoding proteins such as E1 and E2 that are involved in viral replication and transcription regulation. The late region encodes two capsid proteins, L1 and L2. The upstream regulatory region does not encode proteins, and is mainly responsible for regulating the initiation of viral gene transcription and replication.
