Enterovirus 71 (EV71) belongs to the Picornaviridae family, Enterovirus genus, and is a non-enveloped, single-stranded positive-sense RNA virus. Since its initial isolation and identification in 1969, EV71 has become a significant public health pathogen globally, particularly in the Asia-Pacific region. The virus is highly susceptible to infection in the general population, primarily affecting infants and young children. It exhibits strong transmissibility and can be transmitted via the fecal-oral route, respiratory droplets, and close contact. The EV71 viral genome has a relatively high mutation rate. Based on the genetic sequence of its capsid protein VP1, it can be classified into multiple genotypes (A, B, C, D, etc.) and numerous subtypes, with genotypes B and C being the predominant circulating genotypes globally. The continuous evolution of its genome significantly influences the virus's transmissibility and potential antigenic variations.

The Enterovirus 71 virion exhibits a typical icosahedral symmetrical structure, with a diameter of approximately 30 nm. It lacks a lipid envelope, making it highly resistant to environmental factors. The virion structure is highly ordered, primarily consisting of a protein capsid assembled from 60 copies of four structural proteins, which encapsulate the viral genomic RNA. Among these four structural proteins, VP1, VP2, and VP3 are exposed on the surface of the virion, collectively forming the outer shell of the viral capsid. These proteins determine the virus's antigenicity and receptor-binding properties. In contrast, VP4 is located internally within the capsid, interacting with the viral genomic RNA and playing a key role in maintaining capsid stability and the viral uncoating process. A depression on the viral surface, known as the "canyon," serves as the critical site for cellular receptor binding. The viral genomic RNA is approximately 7400 nucleotides in length and contains an open reading frame that encodes a polyprotein precursor. This precursor is processed by virally encoded proteases, ultimately yielding 11 mature proteins, including 4 structural proteins and 7 non-structural proteins. The non-structural proteins are primarily involved in viral genome replication, polyprotein processing, and modulation of host cell physiological functions. High-resolution crystallographic analysis of the EV71 capsid proteins, particularly VP1, has provided crucial molecular insights into its receptor recognition mechanisms, antigenic epitope distribution, and vaccine design.
